Arbetet anv\u00e4nder molekyl\u00e4rcellbiologiska tillv\u00e4gag\u00e5ngss\u00e4tt f\u00f6r att uppt\u00e4cka nya mekanismer som p\u00e5verkar samsjukligheterna av fetma och fettleversjukdom, vars l\u00e5ngsiktiga m\u00e5l \u00e4r att uppt\u00e4cka nya metoder f\u00f6r att f\u00f6rebygga och behandla dessa sjukdomar. <\/p>\n<p style=\"font-weight: 400;\">Det aktuella huvudprojektet handlar om fetma och dess komorbiditeter. Vi har tidigare unders\u00f6kt (i) mekanismerna bakom insulinresistensen och metabolisk dysfunktion hos adipocyter, samt (ii) intracellul\u00e4ra membrankontaktst\u00e4llen (MCS), lipidhomeostas och signalering i prim\u00e4ra endotelceller (EC). F\u00f6r n\u00e4rvarande fokuserar vi p\u00e5 att identifiera defekter i kommunikationen hos adipocyter och EC i fettv\u00e4vnader vid fetma. Vi har optimerat isoleringen av EC och mogna adipocyter fr\u00e5n humana fettbiopsier och etablerat ett samodlingssystem med trans-filter f\u00f6r att observera kommunikationen mellan de tv\u00e5 celltyperna. Vi utf\u00f6r multiomics-analyser av celler isolerade ur fettbiopsier av antingen \u00f6verviktiga eller normalviktiga personer, och utf\u00f6r analyser f\u00f6r att uppt\u00e4cka defekter i adipocyt-EC-kommunikationen vid fetma. M\u00e5let h\u00e4r \u00e4r att uppt\u00e4cka nya behandlingsm\u00e5l som skulle kunna anv\u00e4ndas f\u00f6r att fr\u00e4mja fettv\u00e4vnadsfunktionalitet och f\u00f6r att undvika metaboliska komorbiditeter vid fetma.       <\/p>\n<\/div><div class=\"fusion-text fusion-text-3\"><p style=\"font-weight: 400;\">I ett mer grundl\u00e4ggande forskningsprojekt unders\u00f6ker vi MCS funktioner vid lipidhomeostas och signalering av endotelceller. H\u00e4r har vi utf\u00f6rt systemomfattande analys av utarmningen av endoplasmatiskt retikulum (ER)-Golgi-kontakter, eller av inhibering av lipidtransportproteinet OSBP (oxysterolbindande protein) p\u00e5 dessa platser, vilket avsl\u00f6jar en m\u00e4ngd effekter p\u00e5 lipid\/sterolhomeostas, ER-stress respons och inflammatoriskt genuttryck. F\u00f6r n\u00e4rvarande fokuserar vi p\u00e5 funktionen hos den tidigare d\u00e5ligt studerade OSBP-familjemedlemmen ORP7 i fysiologin f\u00f6r prim\u00e4r EC. Slutm\u00e5let med detta arbete \u00e4r att uppt\u00e4cka EC MCS-funktioner som kan anv\u00e4ndas som m\u00e5l f\u00f6r terapi av kardiometabola sjukdomar som involverar endoteldysfunktion. Ett ytterligare forskningsobjekt i gruppen \u00e4r Golgi-apparatproteinet GOLM1\/GP73, en f\u00f6reslagen biomark\u00f6r f\u00f6r leversjukdomar och flera cancerformer. Funktionen av denna mycket intressanta komponent unders\u00f6ks i cancer\u00f6sa och icke-cancer\u00f6sa hepatocyter och kolangiocyter, varvid syftet \u00e4r att belysa dess potential som ett f\u00f6rmodat terapim\u00e5l f\u00f6r leversjukdomar.
