Olemme kiinnostuneita tyypin 2 diabetekseen (T2D) ja siihen liittyviin piirteisiin vaikuttavista molekulaarisista ja geneettisist\u00e4 mekanismeista. <\/p>\n<p>Olemme kiinnostuneita aineenvaihdunnan s\u00e4\u00e4telyst\u00e4 ja erityisesti insuliinin toimintaa s\u00e4\u00e4televist\u00e4 molekulaarisista mekanismeista ihmisen luurankolihaksessa. K\u00e4yt\u00e4mme t\u00e4rkeimp\u00e4n\u00e4 tutkimusmallina primaareja lihassoluviljelmi\u00e4, jotka on johdettu kliinisesti tarkkaan tutkittujen vapaaehtoisten henkil\u00f6iden lihasbiopsioista. Analysoimme insuliiniresistenssille altistavia ja siihen johtavia mekanismeja sek\u00e4 etsimme uusia tapoja lis\u00e4t\u00e4 insuliiniherkkyytt\u00e4. K\u00e4yt\u00f6ss\u00e4mme on useita erilaisia in vitro -menetelmi\u00e4 solujen aineenvaihdunnan tutkimista varten.   <\/p>\n<p>Finland-United States Investigation of NIDDM Genetics (FUSION) on kansainv\u00e4linen useiden organisaatioiden yhteisty\u00f6projekti, jonka p\u00e4\u00e4tavoitteena on tunnistaa geneettisi\u00e4 riskitekij\u00f6it\u00e4, jotka altistavat tyypin 2 diabetekselle sek\u00e4 siihen liittyvien piirteiden kuten lihavuus, dyslipidemia ja verenpainetauti, kehittymiselle. Ryhm\u00e4johtaja, dosentti Heikki Koistinen, on yksi FUSION-tutkimuksen p\u00e4\u00e4tutkijoista. Esimerkkin\u00e4 tutkimuskysymysten ja tutkimusyhteisty\u00f6n hedelm\u00e4llisest\u00e4 yhdistymisest\u00e4 on p.P50T\/AKT2-geenimuunnoksen tunnistaminen osana FUSION- ja METSIM (Metabolic Syndrome in Men) -tutkimusten meneill\u00e4\u00e4n olevia geneettisi\u00e4 analyyseja. Kyseess\u00e4 on suomalaisille ominainen, insuliiniresistenssille ja tyypin 2 diabetekselle altistava muunnos, joka on hyvin harvinainen ei-suomalaisissa v\u00e4est\u00f6iss\u00e4. Olemme luoneet laajimman ja kansainv\u00e4lisesti ainutlaatuisen primaarien lihassoluviljelmien kokoelman, jonka solulinjat ovat per\u00e4isin suomalaisilta p.P50T\/AKT2 -muunnoksen kantajilta. Olemme my\u00f6s osoittaneet ett\u00e4 insuliinin solunsis\u00e4inen signaalinsiirto on laaja-alaisesti heikentynyt p.P50T\/AKT2 -muunnoksen kantajien lihassoluissa (M\u00e4kinen et al.
