Ty\u00f6ss\u00e4 k\u00e4ytet\u00e4\u00e4n molekyylibiologian l\u00e4hestymistapoja sellaisten uusien mekanismien l\u00f6yt\u00e4miseksi, jotka vaikuttavat lihavuuden ja rasvamaksataudin haittavaikutuksiin. Pitk\u00e4n aikav\u00e4lin tavoite on l\u00f6yt\u00e4\u00e4 uusia l\u00e4hestymistapoja n\u00e4iden sairauksien ehk\u00e4isyyn ja hoitoon. <\/p>\n<p style=\"font-weight: 400;\">Nykyinen p\u00e4\u00e4projekti k\u00e4sittelee lihavuutta ja sen haittavaikutuksia. Olemme tutkineet aiemmin (i) insuliiniresistenssin ja rasvasolujen aineenvaihdunnallisen toimintah\u00e4iri\u00f6n taustalla olevia mekanismeja sek\u00e4 (ii) endoteelisolujen solunsis\u00e4isi\u00e4 kalvokontakteja, rasvojen tasapainoa ja signalointia. Keskitymme t\u00e4ll\u00e4 hetkell\u00e4 lihavan rasvakudoksen rasvasolujen ja endoteelisolujen v\u00e4lisen viestinn\u00e4n virheiden tunnistamiseen. Olemme optimoineet endoteelisolujen ja kypsien rasvasolujen erist\u00e4misen ihmisen rasvakoepaloista sek\u00e4 luoneet yhteisviljelyj\u00e4rjestelm\u00e4n tarkkaillaksemme n\u00e4iden kahden solutyypin v\u00e4list\u00e4 viestint\u00e4\u00e4. Teemme multiomiikka-analyyseja sek\u00e4 lihavien ett\u00e4 normaalipainoisten koehenkil\u00f6iden rasvakudosn\u00e4ytteist\u00e4 eristetyille soluille ja suoritamme testej\u00e4 l\u00f6yt\u00e4\u00e4ksemme vikoja lihavuuteen liittyv\u00e4ss\u00e4 rasvasolujen ja endoteelisolujen v\u00e4lisess\u00e4 viestinn\u00e4ss\u00e4. Tavoitteena on l\u00f6yt\u00e4\u00e4 uusia hoitokohteita, joita voitaisiin hy\u00f6dynt\u00e4\u00e4 rasvakudoksen toiminnan edist\u00e4misess\u00e4 ja lihavuuteen liittyvien aineenvaihdunnallisten haittavaikutusten est\u00e4misess\u00e4.       <\/p>\n<\/div><div class=\"fusion-text fusion-text-3\"><p style=\"font-weight: 400;\">Perustutkimusprojektissa tutkimme solunsis\u00e4isten kalvokontaktien toimintoja endoteelisolujen rasva-ainetasapainossa ja signaloinnissa. Olemme tehneet systeemitason analyysej\u00e4 solulimakalvoston ja Golgin laitteen v\u00e4listen kontaktien v\u00e4hent\u00e4misen tai rasvojen kuljetusproteiini OSBP:n (oksisterolia sitova proteiini) inhibition vaikutuksista n\u00e4iss\u00e4 kohdissa. Tutkimustulokset osoittavat useita vaikutuksia rasva-ainetasapainoon, solulimakalvoston stressivasteisiin ja tulehdusgeenien ilmentymiseen. T\u00e4ll\u00e4 hetkell\u00e4 keskitymme aiemmin heikosti tutkittuun OSBP:n sukuisen ORP7-proteiinin toimintaan endoteelisolujen fysiologiassa. T\u00e4m\u00e4n ty\u00f6n perimm\u00e4inen tavoite on l\u00f6yt\u00e4\u00e4 endoteelisolujen kalvojen kontaktikohtien toimintoja, joita voitaisiin k\u00e4ytt\u00e4\u00e4 endoteelin toimintah\u00e4iri\u00f6ihin liittyvien syd\u00e4n-, verisuoni- ja aineenvaihdunnallisten sairauksien hoidon kohteina. Ryhm\u00e4n lis\u00e4tutkimuskohteena on Golgin laitteen proteiini GOLM1\/GP73, jota on ehdotettu maksasairauksien ja useiden sy\u00f6pien merkkiaineeksi. T\u00e4m\u00e4n kiinnostavan proteiinin toimintaa tutkitaan sy\u00f6p\u00e4- ja ei-sy\u00f6p\u00e4per\u00e4isiss\u00e4 maksan eri solutyypeiss\u00e4. Tarkoituksena on selvitt\u00e4\u00e4 GOLM1:n soveltuvuutta hoitokohteeksi maksasairauksissa.
