Ty\u00f6ss\u00e4 hy\u00f6dynnet\u00e4\u00e4n molekyylisolubiologian l\u00e4hestymistapoja uusien mekanismien l\u00f6yt\u00e4miseksi, jotka vaikuttavat lihavuuden liit\u00e4nn\u00e4issairauksiin. Pitk\u00e4n aikav\u00e4lin tavoitteena on l\u00f6yt\u00e4\u00e4 uusia keinoja n\u00e4iden sairauksien ennaltaehk\u00e4isyyn ja hoitoon.<\/p>\n<p>Nykyinen p\u00e4\u00e4projektimme k\u00e4sittelee lihavuutta ja sen liit\u00e4nn\u00e4issairauksia. Olemme aiemmin tutkineet (i) rasvasolujen (asiposyyttien) insuliiniresistenssin ja metabolisen toimintah\u00e4iri\u00f6n taustalla olevia mekanismeja sek\u00e4 (ii) solunsis\u00e4isi\u00e4 kalvokontaktikohtia (MCS), lipidien homeostaasia ja signalointia primaarisissa endoteelisoluissa (EC). T\u00e4ll\u00e4 hetkell\u00e4 keskitymme tunnistamaan h\u00e4iri\u00f6it\u00e4 rasvasolujen ja endoteelisolujen v\u00e4lisess\u00e4 viestinn\u00e4ss\u00e4 lihavassa rasvakudoksessa. Olemme optimoineet endoteelisolujen ja kypsien rasvasolujen erist\u00e4misen ihmisen rasvabiopsioista ja luoneet suodatinpohjaisen yhteisviljelyj\u00e4rjestelm\u00e4n n\u00e4iden kahden solutyypin v\u00e4lisen viestinn\u00e4n seuraamiseksi. Teemme multiomiikka-analyyseja lihavien tai normaalipainoisten koehenkil\u00f6iden rasvabiopsioista eristetyille soluille ja suoritamme m\u00e4\u00e4rityksi\u00e4 rasvasolujen ja endoteelisolujen v\u00e4lisen viestinn\u00e4n h\u00e4iri\u00f6iden paljastamiseksi lihavuudessa.<\/p>\n<p>Lis\u00e4ksi tutkimme suolistohormonireseptoriagonistin suoria vaikutuksia valkoisen rasvan solutyyppeihin.
