Tutkimusryhm\u00e4mme on hiljattain osoittanut, ett\u00e4 NAFLD:n metabolisten ja geneettisten komponenttien taustalla olevat patogeeniset mekanismit ovat perustavanlaatuisesti erilaisia. Metaboliselle komponentille on tyypillist\u00e4 substraattien ylij\u00e4\u00e4m\u00e4 sek\u00e4 kasvanut rasvakudoksen lipolyysi ja maksan de novo -lipogeneesi (DNL). Sen sijaan geneettiselle komponentille t\u00e4m\u00e4 ei ole tyypillist\u00e4; sit\u00e4 luonnehtii sen sijaan maksan mitokondrioiden kohonnut redokstitila, joka on mitokondrioiden toimintah\u00e4iri\u00f6n merkkiaine.<\/p>\n<p>Tutkimusryhm\u00e4mme on my\u00f6s hiljattain osoittanut, ett\u00e4 PNPLA3-variantti paradoksaalisesti v\u00e4hent\u00e4\u00e4 maksan DNL-synteesi\u00e4, mik\u00e4 liittyi maksan mitokondrioiden sitraattisyntaasin hidastuneeseen vuototasoon (eli sitruunahappokierron nopeudensis\u00e4lt\u00e4v\u00e4\u00e4n vaiheeseen, joka my\u00f6s tuottaa sitraattia DNL-synteesi\u00e4 varten). Sen sijaan ketogeneesi oli lis\u00e4\u00e4ntynyt, koska rasvahappojen hapettumisessa muodostunut asetyyli-Koa ei kyennyt siirtym\u00e4\u00e4n sitruunahappokiertoon. Yhdess\u00e4 n\u00e4m\u00e4 tulokset osoittavat, ett\u00e4 PNPLA3-variantti, joka on NAFLD:n voimakkain geneettinen riskitekij\u00e4, aiheuttaa maksan mitokondrioiden toimintah\u00e4iri\u00f6n.
