Erityisen\u00e4 kiinnostuksen kohteenamme on ei-koodaavien RNA:iden rooli syd\u00e4men vajaatoiminnan kehittymisen ja etenemisen s\u00e4\u00e4telij\u00f6in\u00e4. Tutkimuksemme tavoitteena on l\u00f6yt\u00e4\u00e4 uusia keinoja syd\u00e4men vajaatoiminnan hoitoon ja yksil\u00f6llisemp\u00e4\u00e4n diagnostiikkaan.  <\/p>\n<p>Syd\u00e4n- ja verisuonitaudit ovat t\u00e4rkein sairastuvuuden ja kuolleisuuden syy maailmanlaajuisesti. Syd\u00e4men vajaatoiminta on useiden syd\u00e4n- ja verisuonitautien p\u00e4\u00e4tepiste, ja sen ennuste on edelleen huono optimaalisesta l\u00e4\u00e4kehoidosta huolimatta. Syd\u00e4men vajaatoiminnan taustalla olevan patofysiologian parempi ymm\u00e4rt\u00e4minen on t\u00e4rke\u00e4\u00e4 uusien l\u00e4\u00e4kehoitojen ja diagnostisten ty\u00f6kalujen kehitt\u00e4miseksi.  <\/p>\n<p>Viime vuosikymmenen aikana on osoitettu, ett\u00e4 ei-koodaavilla RNA-molekyyleill\u00e4 (noncoding RNA, ncRNA), kuten mikroRNA:illa (miRNA) ja pitkill\u00e4 ei-koodaavilla RNA:illa (lncRNA) on t\u00e4rke\u00e4 rooli syd\u00e4men vajaatoiminnan patofysiologiassa biologisten prosessien ja signaaliteiden s\u00e4\u00e4telij\u00f6in\u00e4. Tutkimme t\u00e4ll\u00e4 hetkell\u00e4 mekanismeja, joilla miRNA:t ja lncRNA:t s\u00e4\u00e4telev\u00e4t syd\u00e4men vajaatoiminnan kehittymist\u00e4 ja etenemist\u00e4. Tavoitteemme on tunnistaa ja karakterisoida uusia l\u00e4\u00e4kehoidon kohdemolekyylej\u00e4, joiden avulla voitaisiin tehostaa syd\u00e4nlihasvaurion j\u00e4lkeist\u00e4 syd\u00e4men korjaantumista ja regeneraatiota. Uudenlaiset RNA-pohjaiset l\u00e4\u00e4kehoidot voivat tulevaisuudessa parantaa syd\u00e4men vajaatoiminnan ennustetta.   <\/p>\n<p>Tutkimusty\u00f6ss\u00e4mme yhdistyv\u00e4t syd\u00e4nsairauksien kliininen tutkimus ja perustutkimus. K\u00e4yt\u00e4mme mm. syd\u00e4men vajaatoimintaa sairastavien potilaiden syd\u00e4nn\u00e4ytteist\u00e4 tehtyj\u00e4 multiomiikka-analyyseja molekyylitason mekanismien ja uusien l\u00e4\u00e4kehoidon kohdemolekyylien tunnistamiseen. N\u00e4iden kohdemolekyylien toimintaa tutkitaan tarkemmin syd\u00e4nsoluviljelmiss\u00e4 ja seeprakalan syd\u00e4men vajaatoimintamalleissa. Toisin kuin ihmisill\u00e4, joilla syd\u00e4nlihas ei kykene uusiutumaan vaurion j\u00e4lkeen, seeprakalan syd\u00e4n regeneroituu nopeasti vaurion j\u00e4lkeen. Kardiogeeninen sokki on \u00e4killisen syd\u00e4men vajaatoiminnan vakavin muoto. Kehittyneist\u00e4 hoidoista huolimatta sairaalakuolleisuus on edelleen noin 40 %. Uusia merkkiaineita tarvitaan, jotta potilaat voitaisiin paremmin luokitella riskitason perusteella ja hoidon optimointia varten. Aiemmissa tutkimuksissamme mikroRNA:t miR-21-5p, miR-320a-3p ja miR-423-5p olivat itsen\u00e4isi\u00e4 kuolleisuutta ennustavia tekij\u00f6it\u00e4 kardiogeenisess\u00e4 sokissa, mik\u00e4 korostaa ncRNA-molekyylien potentiaalia uusina diagnostisina ja ennusteellisina merkkiaineina.
