Keskitymme proteiinien ubikvitinaatioon sek\u00e4 mitokondrio- ja solulimakalvoston (ER) stressireittien ja autofagian rooliin aivosairauksien patogeneesiss\u00e4 ja sairausmalleissa. Hy\u00f6dynn\u00e4mme tutkimuksessamme erilaisia proteomiikan ja geeninilmentymisen menetelmi\u00e4, soluviljely- ja el\u00e4inmalleja sek\u00e4 geenimuunneltuja hiiri\u00e4. Vaikka suurin osa ryhm\u00e4st\u00e4mme ty\u00f6skentelee l\u00e4\u00e4ketieteellisen tiedekunnan Biomeditsiinin osastolla, ryhm\u00e4 tekee aktiivisesti tutkimusta my\u00f6s Minervassa.<\/p>\n<p>H\u00e4iri\u00f6t proteiinien homeostaasissa liittyv\u00e4t sairauksiin, kuten sy\u00f6p\u00e4\u00e4n ja neurodegeneratiivisiin sairauksiin. Deubikvitinoiva ensyymi USP14 liittyy proteasomiin ja vaikuttaa proteostaasiin (Srinivasan ym., 2023). Usp-14 ilmentyy aivoissa, ja olemme havainneet sen v\u00e4hent\u00e4v\u00e4n mutatoituneen Htt-proteiinin kertymi\u00e4 (Hyrskyluoto ym., 2014). Usp14 voi my\u00f6s vaikuttaa autofagiavuohon autofagiaan liittyv\u00e4n LC3B-proteiinin kautta (Srinivasan ym., 2020). Hy\u00f6dynt\u00e4m\u00e4ll\u00e4 USP14-geenin poistoa olemme \u00e4skett\u00e4in l\u00f6yt\u00e4neet USP14:lle uusia solufysiologisia toimintoja, mukaan lukien vaikutuksia mitokondrioihin ja lysosomeihin, joita tutkimme parhaillaan.<\/p>\n<p>Canopy Homolog 2 (CNPY2) on solulimakalvostossa sijaitseva proteiini, joka osallistuu solujen liikkumiseen, sy\u00f6p\u00e4\u00e4n ja lipoproteiinien aineenvaihduntaan (Do ym., 2012). CNPY2:n yli-ilmentyminen suojaa hermosoluja, kun taas sen ilmentymisen v\u00e4heneminen on niille haitallista. CNPY2 voikin tarjota hy\u00f6ty\u00e4 sairauksissa, joille on tyypillist\u00e4 hermosolujen kato, kuten Huntingtonin taudissa (HD). Tutkimme parhaillaan transgeenist\u00e4 N171-82Q-hiirimallia Huntingtonin taudin mallina k\u00e4ytt\u00e4en erilaisia proteiini- ja molekyylimenetelmi\u00e4 sek\u00e4 k\u00e4ytt\u00e4ytymistestej\u00e4 (Stepanova et.al. 2023).<\/p>\n<p>LACTB on mitokondrion kalvov\u00e4litilan proteiini, joka voi polymeroitua kalvoon kiinnittyviksi kierteisiksi s\u00e4ikeiksi ja muistuttaa bakteerien penisilliini\u00e4 sitovia proteiineja sek\u00e4 beeta-laktamaaseja. LACTB on yhdistetty sy\u00f6p\u00e4\u00e4n kasvainten est\u00e4j\u00e4n\u00e4 (tumorisupressorina) sek\u00e4 lipidiaineenvaihduntaan (Cascone ym., 2022), mutta LACTB:n tarkkoja fysiologisia toimintoja ja mahdollisia substraatteja ei tunneta. T\u00e4ss\u00e4 tutkimuksessa selvit\u00e4mme n\u00e4it\u00e4 kysymyksi\u00e4 pidemm\u00e4lle, mukaan lukien LACTB-poistogeenisten (knock-out) hiirten analyysit.<\/p>\n<\/div><\/div><\/div><div
