Keskitymme proteiinien ubikitinaatioon, mitokondrioiden ja solulimakalvoston stressireittien rooliin sek\u00e4 autofagiaan aivosairauksien patogeneesiss\u00e4 ja malleissa. Hy\u00f6dynn\u00e4mme tutkimuksessa erilaisia proteomisia menetelmi\u00e4 ja geeniekspressiomenetelmi\u00e4, soluviljelmi\u00e4 sek\u00e4 sairauksien el\u00e4inmalleja ja geneettisesti muunneltuja hiiri\u00e4. Vaikka suurin osa ryhm\u00e4st\u00e4mme toimii Medicumissa, ryhm\u00e4 tekee my\u00f6s aktiivisesti tutkimusta Minervassa.  <\/p>\n<p><strong><u>USP14 ja proteiinien ubikitinaatio neurodegeneratiivisissa sairauksissa<\/u><\/strong><\/p>\n<p>H\u00e4iri\u00f6t proteiinien homeostaasissa liittyv\u00e4t sy\u00f6v\u00e4n ja neurodegeneratiivisten sairauksien kaltaisiin tauteihin. Ubikitiini\u00e4 poistava entsyymi USP14 on yhteydess\u00e4 proteasomiin ja vaikuttaa proteostaasiin (Srinivasan et al, 2023). Havaitsimme, ett\u00e4 aivoissa ilmenev\u00e4 USP14 v\u00e4hent\u00e4\u00e4 mutantti-HTT-aggregaatteja (Hyrskyluoto et al, 2014). USP14 voi vaikuttaa my\u00f6s autofagiavirtaukseen autofagiaan liittyv\u00e4n proteiini LC3B:n kautta (Srinivasan et al, 2020). Poistamalla USP14-geenin olemme \u00e4skett\u00e4in havainneet uusia USP14:n toimintoja solufysiologiassa, kuten mitokondrioihin ja lysosomeihin kohdistuvia vaikutuksia, joita tutkitaan t\u00e4ll\u00e4 hetkell\u00e4.    <\/p>\n<p><strong><u>CNPY2 neuroprotektiossa ja solulimakalvoston stressiss\u00e4<\/u><\/strong><\/p>\n<p>Canopy Homolog 2 (CNPY2) on solulimakalvoston proteiini, joka on osallisena solujen liikkuvuudessa, sy\u00f6v\u00e4ss\u00e4 ja lipoproteiinien aineenvaihdunnassa (Do et al, 2012). CNPY2:n yliekspressio suojaa hermosoluja, kun taas vaimennuss\u00e4\u00e4tely on hermosoluille haitallista. CNPY2:sta voi olla jonkinlaista hy\u00f6ty\u00e4 Huntingtonin taudin kaltaisissa sairauksissa, joille on tyypillist\u00e4 hermosolujen tuhoutuminen. Se voi polymerisoitua kalvoon kiinnittyneiksi kierteisiksi s\u00e4ikeiksi, jotka ovat samankaltaisia bakteerien penisilliini\u00e4 sitovien proteiinien ja beetalaktamaasien kanssa. LACTB on yhdistetty sy\u00f6p\u00e4\u00e4n tuumorisuppressorina sek\u00e4 rasva-aineenvaihduntaan (Gascone et al, 2022), mutta LACTB:n tarkkoja fysiologisia toimintoja ja mahdollisia substraatteja ei tunneta. Tutkimme n\u00e4it\u00e4 seikkoja tarkemmin mm.
